Infusion Toxicology
 

The MPI Research Department of Infusion Toxicology has the expertise and technology to conduct intravenous infusion (IV) studies in mice, rats, rabbits, dogs, mini-pigs, and non-human primates. The preclinical safety evaluation of a drug via intravenous infusion is typically used to mimic the route of administration and dose regimen intended for human use.

Difficulties encountered with drugs of poor solubility and/or poor bioavailability when administered via another route can necessitate the use of IV infusion to achieve higher systemic multiples of the clinical dose and/or to control the Cmax value.

The IV infusion allows for larger volumes to be administered over a longer period of time. By adjusting concentration and dose volume, it is possible to increase the tolerability of a drug. The use of a larger blood vessel such as the vena cava also permits a greater dilution of the drug and reduces the potential for irritation due to a low or high pH.

Based on the intended duration of treatment (i.e., from a few minutes up to a continuous 24 hours/day), dosing can be either performed in restrained animals via a peripheral vein or using the jacket tether system. For the latter, a medical-grade catheter is implanted in the vena cava via the femoral vein, or into the jugular vein, then passed subcutaneously and exteriorized in the back of the animal. The catheter is passed through the tether and connected to a rotating swivel attached to the cage, which allows the animal to move freely. On the opposite side of the swivel, short tubing with an injection cap is connected to the syringe or volumetric infusion pump.
